Description | XCL2, also known as lymphotactin-2, is an inflammatory chemokine secreted by T cells and natural killer (NK) cells. XCL2 was initially identified as a variant form of XCL1 that showed only two amino acid differences. XCL1 and XLC2 are structurally similar and share similar agonistic functions. Under physiological conditions, the conformation of XCL1 and XCL2 transitions between the conserved chemokine folds and an unrelated dimeric structure. The monomer form of XCL2 is responsible for the activation of its cognate receptor XCR1, while the dimeric form is important for glycosaminoglycan (GAG) binding. The binding of XCL2 to XCR1 initiates chemotactic cell migration in dendritic cells as well as in T and B lymphocytes. In cancers such as epithelial ovarian and hepatocellular carcinomas, the increased XCL2 expression correlates with cancer progression. Human XCL2 encodes a 114 amino acid precursor protein with a 21 amino acid signal peptide and a single disulfide bond. |
