Description | Purified anti-ADAR1. Adenosine deaminases acting on RNA (ADARs) are involved in RNA editing that converts adenosine residues to inosine in double-stranded RNAs (dsRNAs). The translation machinery reads inosine as guanosine, leading to recoding of a relatively small number of neurotransmitter and ion channel genes and diversification of their functions. Three ADAR gene family members (ADAR1-3) are known in vertebrates. Members of the ADAR gene family share common structural features, such as the presence of multiple dsRNA binding domains (dsRBDs) and a separate deaminase domain. Three repeats of dsRBD are present in ADAR1, while only two repeats are found in ADAR2 and ADAR. ADAR1 increases the rate of pre-microRNA cleavage by Dicer and facilitates loading of miRNA onto RNA-induced silencing complexes. ADAR1 differentiates its functions in RNA editing and RNAi by formation of either ADAR1/ADAR1 homodimer or Dicer/ADAR1 heterodimer complexes. The deficiency of ADAR1 embryonically lethal due to overall apoptosis. |
