JNJ-10198409 is a potent, cell-permeable, ATP-competitive inhibitor of Platelet-Derived Growth Factor receptor tyrosine kinase (PDGF-RTK) with both antiangiogenic and a direct tumor cell antiproliferative activity. It is selective for PDGF-Beta kinase with IC50 values of 4.2 nM for PDGF-Beta and 45 nM for PDGF-a kinase. Displays less activity towards Lck, c-Src, and Fyn while exhibiting little to no inhibition toward VEGFR, HER-2, Cdk's-1,-2,-4 and -7, bFGFR-1 and EGFR. |
