· Sealed and stored at 2-8°C in a cool and dry place.
· Use immediately after unsealing.
· Do not freeze.
· Use immediately once antibacterial agent is dissolved, and it can be stored at 2-8°C for 72h and at -20°C 6 months only after dissolution.
· Human sputum as specimen for the test, which should be digested through alkaline treatment if contaminated.
· Best result will be likely to obtain with first mouthful of sputum spited out of deep lung in the morning.
· Timely detection is recommended after specimen collection, otherwise store at (2-8°C) in the fridge and finish the test within 72 hours.
· Pretreatment of the specimen: alkali treatment and centrifugation
· Take out antibacterial agent, open the cap, and shake up after adding the sterile diluent.
· Suck up 100ul of mixed antibacterial agent, then add into the liquid culture medium and shake up.
· Add the mixed culture medium into the solid culture medium slant.
· Inoculate 0.1ml of already treated specimen onto the slant of the solid culture medium; Do the same thing with the second biphasic L-J medium as the control test.
· Tilt the tube to let the liquid to submerge the entire surface of solid medium, keep the position at 37°C in incubator for 48h, and erect the tube to continue culture.
· Positive: Detection on positive control material
· Negative: Detection on negative control material
